چکیده

The various nucleosides and nucleotides used in this study were commercial preparations. This was also true for inorganic pyrophosphate, tripolyphosphate, and adenosine tetraphosphate. ATP was a crystalline preparation; UTP was further purified by ion exchange chromatography. 1-Pyrophosphoryl ribose 5-phosphate was isolated by the procedure of Kornberg, Lieberman, and Simms (I) to whom we are indebted for a generous sample of this material. Ribose 5-pyrophosphate and ribose 5-triphosphate were synthesized as described else-

Diphosphopyridine nucleotide pyrophosphatase.

Enzymatic splitting of diphosphopyridine nucleotide (DPN)’ by microorganisms, plants, and animal tissues has been known for many years, but the precise nature of this reaction is not well understood (1). Ohlmeyer (2) first observed that adenylic acid (5-phosphoadenosine) appeared when DPN was incubated with yeast maceration juice. Later Heiwinkel (3) identified adenylic acid as a product of the...

Differentiation between ribose-3-phosphate and ribose-5-phosphate by means of the orcinol-pentose reaction.

During a study of the energy-rich phosphorus compounds of higher plants, it became necessary to differentiate between ribose-3-phosphate and ribosed-phosphate. The only method available for distinguishing between these esters at low levels (quantities of 1 mg. or less) is that of Levene and Stiller (1). This method depends on the difference in the rate at which these compounds release their pho...

“coupling” of Phosphorylation with Oxidation of Pyruvic Acid in Brain*

An analysis of the dialyzable components of the pyruvic acid oxidation system of brain showed that in addition to cocarboxylase (thiamine pyrophosphate) (2), a C, dicarboxylic acid (2), and magnesium or manganese ions (3), both inorganic phosphate and “adenine nucleotide” (adenylic acid, adenosine triphosphate) (4) are required. The latter observation suggested that a cycle of phosphorylation i...

Regulation of glucose metabolism by adenine nucleotides in round spermatids from rat testes.

Regulation of glucose metabolism in glycolysis by round spermatids was studied. Assay of activities of 11 glycolytic enzymes in cell-free spermatid extracts showed that hexokinase, phosphofructokinase, and glyceraldehyde-3-phosphate dehydrogenase had the lowest activities. When the cells were incubated with glucose (10 mM), the intracellular level of ATP fell rapidly and 5'-AMP increased. The A...

The Effect of Adenylic Acid on Yeast Nicotinamide Adenine Dinucleotide Isocitrate Dehydrogenase, a Possible Metabolic Control Mechanism.

The nicotinamide adenine dmucleotide-specific isocitrate dehydrogenase of yeast was reported by Kornberg and Pricer (1) to have an absolute requirement for adenylic acid and not to catalyze the reverse reaction under conditions suitable for such catalysis by the nicotinamide adenine dinucleotide phosphatespecific enzyme. Similar results have been obtained for the nicotinamide adenine dinucleoti...
